This route is frequently serviced by a number of bus companies, which is due to the fact that many Slovaks live and work in Prague. You shouldn't have any problems - just turn up - and catch the next bus from Florence Central Station, or even from other smaller stations. Expect to pay around 350 to 500 CZK for a one-way ticket.
